ASAH2B also known as neutral ceramidase B is an enzyme involved in the metabolism of sphingolipids. It has a molecular mass of roughly 100 kDa. The enzyme functions by hydrolyzing ceramides into sphingosine and free fatty acid in a neutral pH environment. ASAH2B is expressed in various tissues including the brain kidney and liver. Its activity contributes significantly to maintaining the balance of sphingolipid-mediated cellular processes.
Biological function summary

ASAH2B impacts cell signaling and microsomal integrity by regulating sphingosine levels. It is not known to be part of a larger complex but it still plays an important role in the ceramide catabolic process. Sphingosine produced by ASAH2B can influence many cellular functions such as apoptosis proliferation and differentiation. It modulates the levels of bioactive lipids impacting cellular homeostasis.

Pathways

The enzyme is integral to the sphingolipid metabolism pathway. By breaking down ceramides ASAH2B facilitates interactions in the sphingolipid signaling pathway which is important for cell fate decisions. ASAH2B works in conjunction with proteins like sphingosine kinase which further converts sphingosine into sphingosine-1-phosphate another signaling molecule essential for cell growth and survival.

ASAH2B dysregulation associates with conditions such as neurodegenerative diseases and cancer. Improper ceramide metabolism influenced by ASAH2B activity may contribute to these pathological states. For instance altered levels of ASAH2B can disrupt normal apoptotic signaling pathways linking it to cancer progression. It also interacts with proteins like ceramide synthase which play roles in these diseases by affecting the balance of pro-apoptotic and pro-growth signals.
