Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

ASCC3 also known as Activating Signal Cointegrator 1 Complex Subunit 3 is a component of the ASC-1 complex involved in transcription regulation and DNA repair activities. It has a mass of around 145 kDa and is expressed in multiple tissues including the brain and other neuronal tissues. This protein plays an essential mechanical role as a helicase which is involved in the unwinding of DNA and RNA influencing how genetic information is read and processed by the cell.
Biological function summary

ASCC3 contributes to cellular responses to DNA damage. It functions as part of the ASC-1 complex which also includes subunits ASCC1 ASCC2 and TRIP4. This complex participates in recruiting other factors to sites requiring repair thereby maintaining genomic stability. The helicase activity of ASCC3 ensures proper transcript processing affects cellular growth and survival under stress conditions.

Pathways

ASCC3 is involved in DNA damage response and repair pathways. Particularly it engages with the nucleotide excision repair (NER) pathway where it affects how cells respond to UV-induced DNA damage. ASCC3 works in conjunction with proteins like XPA and RPA which share roles in recognizing and repairing damaged DNA. This coordination emphasizes ASCC3's relevance to genomic integrity and response to environmental changes.

ASCC3 has been associated with neurological disorders and cancer. Mutations or dysfunctions in ASCC3 can lead to improper DNA repair which increases susceptibility to cancerous transformations. Additionally ASCC3 shows interaction with other proteins related to neuronal health like ASCC1 and mutations in these proteins have been linked to neurodevelopmental disorders. Accurate study of ASCC3's role could help in understanding disease mechanisms and potential therapeutic targets.

Target data

ATPase involved both in DNA repair and rescue of stalled ribosomes (PubMed : 22055184, PubMed : 28757607, PubMed : 32099016, PubMed : 32579943, PubMed : 36302773). 3'-5' DNA helicase involved in repair of alkylated DNA : promotes DNA unwinding to generate single-stranded substrate needed for ALKBH3, enabling ALKBH3 to process alkylated N3-methylcytosine (3mC) within double-stranded regions (PubMed : 22055184). Also involved in activation of the ribosome quality control (RQC) pathway, a pathway that degrades nascent peptide chains during problematic translation (PubMed : 28757607, PubMed : 32099016, PubMed : 32579943, PubMed : 36302773). Drives the splitting of stalled ribosomes that are ubiquitinated in a ZNF598-dependent manner, as part of the ribosome quality control trigger (RQT) complex (PubMed : 28757607, PubMed : 32099016, PubMed : 32579943, PubMed : 36302773). Part of the ASC-1 complex that enhances NF-kappa-B, SRF and AP1 transactivation (PubMed : 12077347).
