Acts as a guanine nucleotide exchange factor (GEF) for RHOA, RAC1 and CDC42 GTPases. Regulates cell migration and adhesion assembly and disassembly through a RAC1, PI3K, RHOA and AKT1-dependent mechanism. Increases both RAC1 and CDC42 activity, but decreases the amount of active RHOA. Required for MMP9 up-regulation via the JNK signaling pathway in colorectal tumor cells. Involved in tumor angiogenesis and may play a role in intestinal adenoma formation and tumor progression.

Asef2 also known as SPATA13 is a Rac-specific guanine nucleotide exchange factor (GEF) that primarily influences actin cytoskeleton dynamics. It has a mass of approximately 91 kDa and contains a DH-PH domain arrangement. Asef2 is expressed in various tissues including the brain kidney and testis. Its role involves modulating the activity of small GTPases particularly Rac1 which are critical for cellular morphological adjustments.
In the context of cellular dynamics Asef2 facilitates the reorganization of the cytoskeleton by promoting actin filament assembly and stability. It acts within the context of protein complexes that regulate cell migration and adhesion.
