ASF1A and ASF1B also known as Anti-silencing Function 1A and 1B are histone chaperones. They both have a molecular mass around 22 kDa and play an important role in regulating nucleosome assembly by facilitating histone H3-H4 dimer transfer. These proteins are usually expressed in the nucleus of dividing cells and are present in nearly all eukaryotic organisms. They maintain genomic stability and their activity is tightly regulated due to their involvement in the cell cycle.
Biological function summary

ASF1A and ASF1B serve important roles by assisting in DNA replication and repair. They participate in chromatin remodeling and transcriptional regulation through their interaction with other proteins and chromatin complexes. ASF1A and ASF1B function closely with histone H3-H4 tetramers ensuring proper histone storage and supply during S-phase of cell cycle. Their interaction with complexes like the FACT complex helps maintain a dynamic chromatin structure.

Pathways

The histone chaperones ASF1A and ASF1B are integrated into DNA replication and repair pathways. They associate with the DNA damage response pathway as they interact with the ATR/ATM kinase signaling which is important for repairing DNA double-strand breaks. ASF1 proteins also work with chromatin assembly factors to assist the synthesis and stabilization of replication forks. These pathways connect closely with other proteins like CAF-1 which also helps in chromatin assembly during DNA replication.

Disruptions in ASF1A and ASF1B functions have links to cancer and neurodegenerative disorders. Overexpression of ASF1A is observed in certain types of cancer suggesting a role in tumorigenesis through interactions with proteins such as histone H3. Mutations in ASF1B can contribute to neurodegenerative disorders by affecting chromatin structure and function. These disturbances highlight their significant roles and potential as therapeutic targets.

Target data

Histone chaperone that facilitates histone deposition and histone exchange and removal during nucleosome assembly and disassembly (PubMed : 10591219, PubMed : 12381660, PubMed : 14561777, PubMed : 16151251, PubMed : 16396992). Cooperates with chromatin assembly factor 1 (CAF-1) to promote replication-dependent chromatin assembly (PubMed : 11533245). Plays a role in the formation of silent heterochromatin (PubMed : 12381660, PubMed : 28320872).
