ASIC2 also known as amiloride-sensitive cation channel 2 or ACCN1 is a protein involved in ion transport. It forms part of the acid-sensing ion channel (ASIC) family and functions as a voltage-insensitive proton-gated ion channel. ASIC2 is known to have a mass of approximately 60 kDa. Expression is mainly noted in the central nervous system particularly in neurons as well as in other tissues such as the heart and lungs.
Biological function summary

ASIC2 contributes to the detection and response to changes in extracellular pH playing a role in processes such as pain perception and synaptic plasticity. ASIC2 can form homo- or heteromeric complexes with other ASIC proteins influencing the channel properties like ion selectivity and gating. By participating in these complexes ASIC2 affects neuronal signaling and potentially mood regulation in the central nervous system.

Pathways

ASIC2 plays a role in the proton-activated pathways by influencing neuronal excitability. One key pathway includes the pain transduction pathway where ASIC2 in combination with other ASIC family members like ASIC1 and ASIC3 helps mediate pain sensation in response to acids. This regulation is important for maintaining normal sensory functions and could be linked with synaptic functions and neurodegenerative processes.

ASIC2 relates to conditions such as ischemic brain injury and anxiety disorders. During ischemic events ASIC2 contributes to neuronal death making it a target for potential therapeutic interventions. In anxiety disorders alterations in ASIC2 expression or function could impact mood regulation. The protein interacts with other proteins like ASIC1 in these disorder-related pathways suggesting that modulating ASIC interactions might offer a strategy for therapeutic developments.
