ATF7 (activating transcription factor 7) is a transcription factor that belongs to the ATF/CREB family. It has alternative names such as ATFa and B-ATF. Mechanically ATF7 regulates transcription by binding to cyclic AMP response elements (CRE) in DNA. This protein has a mass of approximately 55 kDa. ATF7 is expressed in various tissues throughout the body including the brain liver and spleen supporting its involvement in multiple physiological processes.
Biological function summary

ATF7 functions in cellular stress responses and cell differentiation. It often forms a complex with other transcription factors to exert its effects on gene expression. ATF7 is known to influence the expression of genes involved in stress response pathways and apoptosis. By regulating these genes ATF7 plays a role in how cells react to environmental changes and maintain homeostasis.

Pathways

ATF7 is involved in significant signaling pathways such as the MAPK and JNK pathways. These pathways are important for transmitting signals from the cell surface to the nucleus affecting gene expression. ATF7 interacts with proteins such as c-Jun and ATF2 within these pathways facilitating cellular responses to stress and growth signals.

ATF7 has been linked to certain cancers and neurodegenerative diseases. Abnormal expression or mutation of ATF7 can contribute to the progression of cancer possibly through its interaction with proteins like c-Myc. In neurodegenerative conditions ATF7 might play a role in neuronal cell death potentially in connection with proteins involved in stress signaling such as p53. Understanding the role of ATF7 in these disorders may provide insight into new therapeutic strategies.

Target data

Stress-responsive chromatin regulator that plays a role in various biological processes including innate immunological memory, adipocyte differentiation or telomerase regulation (PubMed : 29490055). In absence of stress, contributes to the formation of heterochromatin and heterochromatin-like structure by recruiting histone H3K9 tri- and di-methyltransferases thus silencing the transcription of target genes such as STAT1 in adipocytes, or genes involved in innate immunity in macrophages and adipocytes (By similarity). Stress induces ATF7 phosphorylation that disrupts interactions with histone methyltransferase and enhances the association with coactivators containing histone acetyltransferase and/or histone demethylase, leading to disruption of the heterochromatin-like structure and subsequently transcriptional activation (By similarity). In response to TNF-alpha, which is induced by various stresses, phosphorylated ATF7 and telomerase are released from telomeres leading to telomere shortening (PubMed : 29490055). Also plays a role in maintaining epithelial regenerative capacity and protecting against cell death during intestinal epithelial damage and repair (By similarity).. Isoform 4. Acts as a dominant repressor of the E-selectin/NF-ELAM1/delta-A promoter.. Isoform 5. Acts as a negative regulator, inhibiting both ATF2 and ATF7 transcriptional activities. It may exert these effects by sequestrating in the cytoplasm the Thr-53 phosphorylating kinase, preventing activation.
