The ATP12A protein also known as gastric H+/K+ ATPase functions as a proton pump in cellular processes. It facilitates the exchange of potassium ions (K+) and hydrogen ions (H+) across cell membranes playing an essential role in ion homeostasis. The ATP12A protein has a molecular mass of approximately 116 kDa. Expression of ATP12A is found primarily in the gastric epithelium where it contributes to the acidification of the stomach although it can also be present in other tissues such as the colon.
Biological function summary

Ion transport is significantly influenced by ATP12A. This protein reforms a part of an enzyme complex that provides energy for H+ ion secretion. By coupling ATP hydrolysis to the transport process ATP12A catalyzes reactions vital for maintaining the acidic environment in the stomach. The protein's activity relates closely with potassium channels with which it might interact to optimize gastric acid secretion.

Pathways

ATP12A is integral to the gastric acid secretion pathway. Through this pathway the protein collaborates with the neighboring potassium chloride transporters to regulate acid release in the gastric lumen. ATP12A also shares a functional relationship with ATP1AL1 another ATPase variant in facilitating ATP-dependent ion transport. These pathways and interactions highlight the protein's central role in maintaining cellular processes related to pH balance.

ATP12A is linked with certain gastric pathologies such as peptic ulcers and gastritis. The protein's role in abnormal gastric acid secretion can exacerbate these conditions highlighting its impact on gastrointestinal health. Malfunction in related proteins such as ATP4A can further influence the pathological outcomes associated with ATP12A activity leading to exacerbated symptoms of gastric dysfunctions.

The catalytic subunit of a H(+)/K(+) ATPase and/or Na(+)/K(+) ATPase pump which transports K(+) ions in exchange for Na(+) and/or H(+) ions across the apical membrane of epithelial cells. Uses ATP as an energy source to pump K(+) ions into the cell while transporting Na(+) and/or H(+) ions to the extracellular compartment (PubMed : 11341842, PubMed : 7485470, PubMed : 8853415, PubMed : 9774385). Involved in the maintenance of electrolyte homeostasis through K(+) ion absorption in kidney and colon (By similarity). In the airway epithelium, may play a primary role in mucus acidification regulating its viscosity and clearance (PubMed : 29391451).
