ATP1A2 also known as the Na+/K+ ATPase alpha-2 subunit is integral in the maintenance of the sodium and potassium gradient across the plasma membrane. This protein with a mass around 113 kDa functions as part of the Na+/K+ ATPase pump and is essential for cellular ion homeostasis. ATP1A2 is expressed in various tissues with high expression noted in the brain heart and skeletal muscle. The pump activity provides necessary conditions for maintaining resting membrane potential and supporting cellular processes such as nutrient transport.
Biological function summary

ATP1A2 contributes to neuronal excitability and muscle contraction. It forms a part of the Na+/K+ ATPase enzyme complex which plays a significant role in neurotransmitter release and action potential propagation in neurons. This action is critical in both central and peripheral nervous systems. The interaction of ATP1A2 with intracellular signaling pathways also influences cardiac muscle function impacting heart rate and contractility.

Pathways

ATP1A2 plays an essential role in the regulation of certain vital biochemical pathways like the electrical excitability in nerve cells and the cardiac action potential pathways. These pathways coordinate a variety of physiological responses and maintain cellular energetics. The protein pathways involving ATP1A2 often intersect with those of other ion channels and transporters such as the NCX (sodium-calcium exchanger) which further modulate cellular ion exchange and signaling processes.

ATP1A2 mutations have associations with conditions such as familial hemiplegic migraine type 2 and alternating hemiplegia of childhood. These disorders often result from disrupted ion homeostasis leading to abnormal neural activity. The altered function of ATP1A2 in these diseases may also show interaction with proteins involved in neurotransmitter regulation such as those involved in the glutamatergic and GABAergic signaling pathways potentially exacerbating neural dysfunction and symptoms.
