ATP6V0A1 also known as a subunit of the V0 domain of the vacuolar ATPase (V-ATPase) is a transmembrane protein involved in the acidification of intracellular compartments. It forms a component of the proton channel within the V-ATPase. This protein weighs approximately 95 kDa and can be found expressed in many tissues such as the kidney brain and liver. Its mechanical role involves contributing to proton translocation across cell membranes which is essential for various cellular activities.
Biological function summary

The protein participates in the ATP-dependent acidification of organelles like lysosomes and endosomes. As a part of a multi-subunit V-ATPase complex ATP6V0A1 helps maintain cellular pH balance and is essential for processes such as protein degradation and receptor-mediated endocytosis. Its activity influences the function of numerous cellular processes including ion transport and neurotransmitter release by regulating the pH in synaptic vesicles.

Subunit of the V0 complex of vacuolar(H+)-ATPase (V-ATPase), a multisubunit enzyme composed of a peripheral complex (V1) that hydrolyzes ATP and a membrane integral complex (V0) that transports protons across cellular membranes. V-ATPase is responsible for the acidification of various organelles, such as lysosomes, endosomes, the trans-Golgi network, and secretory granules, including synaptic vesicles (PubMed : 33065002, PubMed : 33833240, PubMed : 34909687). In certain cell types, can be exported to the plasma membrane, where it is involved in the acidification of the extracellular environment (By similarity). Required for assembly and activity of the vacuolar ATPase (By similarity). Through its action on compartment acidification, plays an essential role in neuronal development in terms of integrity and connectivity of neurons (PubMed : 33833240).
