The B27 protein also known as HLA-B27 is a specific allele of the human leukocyte antigen (HLA) class I molecules. This protein has a mass of approximately 45 kDa and is mainly expressed on the surface of nucleated cells especially in immune cells like lymphocytes. HLA-B27 plays an important role in presenting peptides to the immune system enabling the recognition and clearance of pathogens. This expression helps maintain immune surveillance and promotes the initiation of immune responses.
Biological function summary

This protein assists in the regulation of immune system activities. HLA-B27 does so by forming a complex with beta-2 microglobulin and presenting endogenous antigens to CD8+ T cells. This presentation is important for immune regulation and cellular immunity. The ability of HLA-B27 to bind peptides often shapes immunological responses influencing how the immune system reacts to infectious agents and other antigens.

Pathways

This protein significantly integrates into the antigen processing and presentation pathway. It contributes to the adaptive immune response playing a role in the endosomal/lysosomal pathway where intracellular proteins get degraded into peptides. HLA-B27 works closely with other HLA molecules and the transporter associated with antigen processing (TAP) facilitating efficient peptide loading and transport onto the cell surface.

HLA-B27 shows a strong association with several autoimmune conditions including ankylosing spondylitis and reactive arthritis. Genetic studies suggest that misfolded HLA-B27 may lead to endoplasmic reticulum stress influencing cellular processes related to these conditions. Tumor necrosis factor-alpha (TNF-alpha) often interacts with HLA-B27 within these disease contexts which further highlights the protein's involvement in inflammatory pathways and autoimmune manifestations.

Modulator of melanocortin receptors (MC1R, MC2R, MC3R, MC4R and MC5R). Acts by increasing ligand-sensitivity of melanocortin receptors and enhancing generation of cAMP by the receptors. Required both for MC2R trafficking to the cell surface of adrenal cells and for signaling in response to corticotropin (ACTH). May be involved in the intracellular trafficking pathways in adipocyte cells.
