The B3GAT1 protein also known as CD57 or HNK-1 functions mechanically as a β-13-glucuronyltransferase. It has a mass of approximately 55 kDa. B3GAT1 localizes mainly in the Golgi apparatus and is predominantly expressed in neural and immune tissues. The protein plays an important role in the biosynthesis of the HNK-1 carbohydrate epitope which is significant for cellular communication and molecular interactions in the nervous system.
Biological function summary

B3GAT1 participates in the generation of specific carbohydrate structures that contribute to cell-cell adhesion processes. It is a part of a larger complex of glycosyltransferases that include enzymes like β3GalT1 and ST3GalT6 which together help build complex glycan chains. By modifying glycoproteins and glycolipids B3GAT1 influences neural connectivity and immune response modulation.

Pathways

B3GAT1 influences with its role in the glycosphingolipid biosynthesis pathway and the neural crest cell migration pathway. In glycosphingolipid biosynthesis its activity is essential for creating carbohydrate polymer structures linking it with proteins like β3GalT2. It also interacts with NCAM1 within the neural crest cell migration pathway impacting neural development and plasticity.

Abnormal expression or mutations in B3GAT1 link to diseases such as Alzheimer's as the HNK-1 epitope expression correlates with neuronal impairment. In cancers altered B3GAT1 glycosylation patterns associate with tumor progression and immune escape mechanisms where its interaction with CD56 plays a role in modulating immune responses.

Target data

Involved in the biosynthesis of L2/HNK-1 carbohydrate epitope on glycoproteins. Can also play a role in glycosaminoglycan biosynthesis. Substrates include asialo-orosomucoid (ASOR), asialo-fetuin, and asialo-neural cell adhesion molecule. Requires sphingomyelin for activity : stearoyl-sphingomyelin was the most effective, followed by palmitoyl-sphingomyelin and lignoceroyl-sphingomyelin. Activity was demonstrated only for sphingomyelin with a saturated fatty acid and not for that with an unsaturated fatty acid, regardless of the length of the acyl group.
