Supplementary information

B3GNT1 also known as UDP-GlcNAc:betaGal beta-13-N-acetylglucosaminyltransferase 1 is an enzyme that participates in glycosylation processes by transferring N-acetylglucosamine from UDP-GlcNAc to polylactosamine chains playing an important role in the synthesis of poly-N-acetyllactosamine. This protein has a mass of approximately 43 kDa and predominantly expresses in tissues that produce substantial amounts of glycosylated molecules such as the brain and the gastrointestinal tract.
Biological function summary

B3GNT1 contributes to the synthesis of glycan structures that are important for cell-cell interactions. It functions as an independent enzyme rather than as part of a larger enzymatic complex. The activity of B3GNT1 impacts the production of complex-type N- and O-glycans which support cellular adhesion signaling and immune recognition. Such functions make B3GNT1 significant in various physiological processes particularly in the development and maintenance of tissues.

Pathways

B3GNT1 is integral to the glycosaminoglycan biosynthesis and glycoprotein metabolic pathways. It interacts with other enzymes involved in these pathways such as beta-14-galactosyltransferase to construct poly-N-acetyllactosamine structures. The connectivity between B3GNT1 and other glycosyltransferases ensures proper assembly and function of glycan-containing molecules necessary for diverse biological activities.

Aberrant expression or malfunction of B3GNT1 links to conditions like congenital disorders of glycosylation and certain cancers. Abnormal glycosylation patterns sometimes involving related proteins such as FUT8 which is another glycosyltransferase can lead to defective cellular signaling and contribute to these pathologies. Understanding these connections helps in exploring therapeutic targets and diagnostic markers for glycosylation-related diseases.

Target data

Beta-1,3-N-acetylglucosaminyltransferase involved in the synthesis of poly-N-acetyllactosamine. Catalyzes the initiation and elongation of poly-N-acetyllactosamine chains. Shows a marked preference for Gal(beta1-4)Glc(NAc)-based acceptors (PubMed : 9892646). Probably constitutes the main polylactosamine synthase.
