Supplementary information

B3GNT7 also known as β-13-N-acetylglucosaminyltransferase 7 is an enzyme involved in glycosylation processes. It has a mass of approximately 46 kDa. Found within the Golgi apparatus of cells B3GNT7 is responsible for catalyzing the transfer of N-acetylglucosamine (GlcNAc) residues to precursor oligosaccharides forming β1-3 glycosidic bonds. It shows widespread expression but levels are higher in tissues such as brain pancreas and kidneys indicating tissue-specific roles.
Biological function summary

B3GNT7 plays an important role in the biosynthesis of glycosaminoglycans which are complex polysaccharides. These polysaccharides provide structure and support to cells and tissues partaking in processes like cell signaling and maintaining cellular environment. B3GNT7 is not part of multienzyme complexes but interacts closely with other glycosylation enzymes. This specificity influences the structural diversity and function of glycosaminoglycans in various biological processes.

Pathways

B3GNT7 participates in the glycosaminoglycan biosynthesis pathway particularly in the synthesis of keratan sulfate. This pathway impacts cellular communication and structural integrity significant for tissue function. B3GNT7 functions alongside related enzymes such as B3GNT1 which also contribute to glycosaminoglycan chain elongation. The interactions with these enzymes play an important role in modifying glycosaminoglycan chains for distinct biological functions.

Alterations in B3GNT7 expression and function relate to osteoarthritis and certain neuropathies. In osteoarthritis abnormal glycosaminoglycan synthesis can lead to impaired joint function and cartilage deterioration. Interactions with proteins like B3GNT6 which has overlapping roles in glycosylation may influence disease progression. Understanding B3GNT7's impact on these conditions helps in developing targeted therapeutic strategies.

Target data

N-acetyl glucosamine (GlcNAc) transferase that catalyzes the transfer of GlcNAc via a beta1->3 linkage from UDP-GlcNAc to the non-reducing terminal galactose (Gal) in the linearly growing chain of N- and O-linked keratan sulfate proteoglycans. Cooperates with B4GALT4 galactosyltransferase and CHST6 and CHST1 sulfotransferases to construct and elongate mono- and disulfated disaccharide units [->3Galbeta1->4(6-sulfoGlcNAcbeta)1->] and [->3(6-sulfoGalbeta)1->4(6-sulfoGlcNAcbeta)1->] within keratan sulfate polymer (PubMed : 14706853, PubMed : 17690104). Involved in biosynthesis of N-linked keratan sulfate proteoglycans in cornea, with an impact on proteoglycan fibril organization and corneal transparency (By similarity) (PubMed : 17690104). May play a role in the maintenance of tissue architecture by suppressing cellular motility and invasion (By similarity).
