B4Gal-T6 also known as beta-14-galactosyltransferase 6 is an enzyme that catalyzes the transfer of galactose to specific acceptor molecules like N-acetylglucosamine. It is approximately 46 kDa in size. It plays a role in the biosynthesis of glycoproteins and glycolipids. This enzyme is expressed in several tissues including the liver kidney and brain. Its activity contributes to the formation of complex carbohydrates that occur on cell surfaces and in secreted proteins.
B4Gal-T6 functions in the assembly of glycosaminoglycans (GAGs) which form the carbohydrate components of the extracellular matrix. This enzyme acts as part of a larger complex involving other glycosyltransferases that regulate the elongation of polysaccharide chains. It ensures the proper structure and function of proteoglycans in tissues affecting cell adhesion migration and proliferation.
B4Gal-T6 is involved in the glycosylation pathways related to the synthesis of GAGs and the N-linked glycosylation of proteins. It interacts with other enzymes like b4Gal-T1 within these pathways ensuring the correct formation and modification of glycan structures. The enzyme contributes to pathways such as the Wnt signaling pathway where proper glycosylation is necessary for pathway activation and function.
Abnormal b4Gal-T6 activity can associate with cancer and Ehlers-Danlos syndrome. In cancer altered glycosylation patterns can influence tumor growth and metastasis. The enzyme interacts with proteins such as decorin which is involved in collagen fibril organization and skin integrity linking its dysfunction to Ehlers-Danlos syndrome.
