B4GALNT1 also known as beta-14-N-acetylgalactosaminyltransferase 1 is an enzyme that plays a role in the biosynthesis of gangliosides. It has a molecular mass of approximately 56 kDa. B4GALNT1 catalyzes the transfer of N-acetylgalactosamine to substrates in the Golgi apparatus which facilitates the formation of gangliosides. This enzyme exhibits expression mainly in the central nervous system and several other tissues reflecting its role in the development and function of neural cells.
The enzyme contributes to the synthesis of complex gangliosides. B4GALNT1 does not work alone; instead it integrates into a system of glycosylation pathways involving other glycosyltransferases forming a complex network that tailors the composition of glycan structures on lipids and proteins. This activity influences cell recognition adhesion and signaling processes important for neurodevelopment and synapse formation.
B4GALNT1 influences the ganglioside biosynthetic pathway an important aspect for proper neuronal function. This pathway involves collaboration with other proteins such as GM2 synthase which cooperates in the addition of sugar residues at various stages of glycolipid formation. These interactions ensure the synthesis of diverse ganglioside molecules within nerve cell membranes which are vital in modulating cellular interactions and signal transduction.
Alterations in B4GALNT1 function have connections to neurological disorders such as Alzheimer's disease and certain types of gangliosidosis. Perturbations in its enzymatic activity can disrupt normal ganglioside composition affecting membrane dynamics and cell signaling. Phenomena linked to other glycosylation enzymes like GM3 synthase also illustrate disease pathways impacted by aberrant glycan synthesis highlighting B4GALNT1's indirect influence on these disorders.

Low expression: no staining on human skeletal muscle. The section was incubated with ab314130 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.
