Supplementary information

B4GALT5 also known as beta-14-galactosyltransferase 5 is an enzyme critical for glycosylation processes. This enzyme has a molecular weight of approximately 43 kDa. It adds galactose to specific carbohydrate structures and plays a role in forming lactose and other glycan structures. This enzyme is part of the larger beta-14-galactosyltransferase family and is found in the Golgi apparatus. It is expressed in various tissues with significant activity in the liver and kidney and to a certain extent in the brain.
Biological function summary

B4GALT5 is involved in the synthesis of complex glycoconjugates and lactose. It contributes to the N-linked and O-linked glycosylation of proteins which are essential for protein folding stability and cell signaling. B4GALT5 forms part of the enzyme complex responsible for elongating glycan chains cooperating with other glycosyltransferases. Through its enzymatic activity it affects cell-cell interactions host-pathogen recognition and immune responses.

Pathways

B4GALT5 is an important component of the glycan biosynthesis and metabolism pathways. Its action affects the glycoprotein quality control and folding pathways. In these processes it works alongside other glycosyltransferases such as B4GALT1 ensuring proper glycoconjugate formation and modification. These pathways influence developmental processes immune system functions and normal cellular operations.

B4GALT5 has links to congenital disorders of glycosylation and cancer. Abnormal glycosylation can alter cell adhesion and migration contributing to tumorigenesis and metastasis. The enzyme shows altered expression levels in certain cancers suggesting its role as a potential biomarker. B4GALT5 has associations with other proteins like E-cadherin where changes in glycosylation patterns can influence cancer progression and immune evasion.

Target data

Catalyzes the synthesis of lactosylceramide (LacCer) via the transfer of galactose from UDP-galactose to glucosylceramide (GlcCer) (PubMed : 24498430). LacCer is the starting point in the biosynthesis of all gangliosides (membrane-bound glycosphingolipids) which play pivotal roles in the CNS including neuronal maturation and axonal and myelin formation (By similarity). Plays a role in the glycosylation of BMPR1A and regulation of its protein stability (By similarity). Essential for extraembryonic development during early embryogenesis (By similarity).
