Bad pSer75 also known as Bcl-2-associated death promoter Ser75 is a phosphorylation site of the BAD protein an important pro-apoptotic member of the Bcl-2 family. BAD is a protein that undergoes phosphorylation to regulate its function. Its mass is about 18 to 20 kDa and it expresses in various tissues including the brain heart and immune cells. The phosphorylation at serine 75 plays an important role in modulating BAD's interaction with other proteins.
Biological function summary

When phosphorylated at serine 75 BAD associates with 14-3-3 proteins which sequesters it in the cytoplasm and inhibits its pro-apoptotic activity. This modification is part of a larger signaling complex that influences cell survival. Phosphorylation of BAD affects its ability to bind to and neutralize anti-apoptotic proteins like Bcl-xL and Bcl-2 critical in balancing cell death and survival.

Pathways

BAD pSer75 phosphorylation integrates into survival pathways especially the PI3K/Akt and MAPK/ERK pathways. Akt and ERK kinases regulate this phosphorylation site and promote cell survival by inactivating BAD. These pathways interact with other players like mTOR and promote cellular responses to growth signals delineating a critical role in interpreting extracellular cues that affect apoptosis.

Dysregulation of BAD pSer75 phosphorylation connects to cancer and neurodegenerative disorders. In cancers abnormal phosphorylation could alter cell survival aiding the cancer cells’ resistance to apoptosis. BAD relates to proteins like Bcl-2 which also play roles in cancer pathways. In neurodegenerative diseases improper regulation of apoptosis contributes to cellular loss. Changes in BAD activity influenced through its phosphorylation contribute to the disease state by failing to maintain proper neuronal survival.

Promotes cell death. Successfully competes for the binding to Bcl-X(L), Bcl-2 and Bcl-W, thereby affecting the level of heterodimerization of these proteins with BAX. Can reverse the death repressor activity of Bcl-X(L), but not that of Bcl-2 (By similarity). Appears to act as a link between growth factor receptor signaling and the apoptotic pathways.
