Involved in transcriptional activation and repression of select genes by chromatin remodeling (alteration of DNA-nucleosome topology). Component of SWI/SNF chromatin remodeling complexes that carry out key enzymatic activities, changing chromatin structure by altering DNA-histone contacts within a nucleosome in an ATP-dependent manner. Required for maximal ATPase activity of SMARCA4/BRG1/BAF190A and for association of the SMARCA4/BRG1/BAF190A containing remodeling complex BAF with chromatin/nuclear matrix. Belongs to the neural progenitors-specific chromatin remodeling complex (npBAF complex) and is required for the proliferation of neural progenitors. During neural development a switch from a stem/progenitor to a postmitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. The transition from proliferating neural stem/progenitor cells to postmitotic neurons requires a switch in subunit composition of the npBAF and nBAF complexes. As neural progenitors exit mitosis and differentiate into neurons, npBAF complexes which contain ACTL6A/BAF53A and PHF10/BAF45A, are exchanged for homologous alternative ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C subunits in neuron-specific complexes (nBAF). The npBAF complex is essential for the self-renewal/proliferative capacity of the multipotent neural stem cells. The nBAF complex along with CREST plays a role regulating the activity of genes essential for dendrite growth (By similarity). Component of the NuA4 histone acetyltransferase (HAT) complex which is involved in transcriptional activation of select genes principally by acetylation of nucleosomal histones H4 and H2A. This modification may both alter nucleosome - DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. This complex may be required for the activation of transcriptional programs associated with oncogene and proto-oncogene mediated growth induction, tumor suppressor mediated growth arrest and replicative senescence, apoptosis, and DNA repair. NuA4 may also play a direct role in DNA repair when recruited to sites of DNA damage. Putative core component of the chromatin remodeling INO80 complex which is involved in transcriptional regulation, DNA replication and probably DNA repair.

In Western blot, ab3882 detects BAF53 and Tip60-TAP in total extracts of Tip60-TAP. Following TEV elution of Tip60-TAP ab3882 only detects BAF53. The protein BAF53A also known as ACTL6A plays an important role in gene expression through chromatin remodeling. It is a subunit of the SWI/SNF-like BAF complex that is responsible for altering chromatin structure making it easier for transcription factors to access DNA. BAF53A has a molecular mass of approximately 53 kDa. It is highly expressed in neurons and plays an essential role in neurodevelopment and the regulation of neuronal gene expression.
BAF53A is an integral component of the BAF chromatin remodeling complex. This complex modifies the positioning and accessibility of nucleosomes on chromatin thereby influencing gene transcription processes. BAF53A contributes to the regulation of genes involved in neuronal differentiation and development. The protein plays a significant role in neural stem cell proliferation and the regulation of progenitor cell fate.
The BAF53A protein participates actively in the neural development and chromatin remodeling pathways. It interacts closely with the BAF complex to regulate the transcription of genes essential for neural differentiation and development. The BAF complex which includes BAF53A associates with proteins such as BRG1 and SMARCC1 facilitating the unfolding of chromatin to affect gene expression positively.
BAF53A is associated with neurodevelopmental disorders and certain types of cancer. Altered expression or mutations in BAF53A have been linked to neurodevelopmental issues contributing to conditions such as Autism Spectrum Disorders. Additionally overexpression of BAF53A connects to cancer particularly in glioblastoma where it influences tumor progression. In these contexts BAF53A interacts with oncogenes and tumor suppressor proteins playing a role in disease pathogenesis.

Sonicated Chromatin prepared from untreated (UI) or 17beta-estradiol (E2) treated MCF7 cells was subjected to the ChIP procedure with ab3882 to BAF53A and the immunoprecipitated chromatin was analysed in the proximal region of the estrogen-responsive pS2 promoter (as shown above) and quantified by real-time PCR (values are nomalized over inputs). The primers are designed to follow the nucleosome E (including the Estrogen Responsive Element ERE). 8 μl of ab3882 and 2x106 cells were used in each ChIP experiment.
Total extracts from two different clones of MCF7 (Human breast adenocarcinoma cell line) cells expressing a TAP tagged version of Tip60 were partially purified over IgG sepharose resin and eluted with TEV protease. Western blot was performed using a 1/1000 dilution of the BAF53A antibody.
