Supplementary information

BarX1 also known as BarH-like homeobox 1 is a transcription factor with a molecular mass of approximately 29 kDa. It is part of the Bar family of proteins which play essential roles in development. BarX1's primary expression occurs in the central nervous system and mesenchymal tissues during embryonic development. In adults BarX1 shows limited expression in specific tissues such as the stomach where it may serve various regulatory functions.
Biological function summary

BarX1 functions in the development of multiple organ systems particularly in teeth and stomach. It acts by influencing the differentiation and proliferation of cells during the formation of these structures. BarX1 is known to be part of a regulatory complex that controls the expression of genes critical for morphogenesis and tissue specification. Its interaction with other homeobox proteins further expands its impact on development.

Pathways

BarX1 interacts in key developmental signaling pathways such as Wnt and Hedgehog. These pathways play significant roles in development influencing cell fate and patterning. BarX1's interaction with proteins like beta-Catenin in the Wnt pathway helps modulate the activity of downstream genes necessary for development. By participating in these pathways BarX1 helps maintain the balance between cell proliferation and differentiation.

Dysregulation of BarX1 expression has been linked to conditions such as gastric cancer and tooth agenesis. In gastric cancer abnormal BarX1 levels can disrupt normal gene expression leading to uncontrolled cell growth and tumor development. Connections also exist between BarX1 and E-cadherin; alterations in this interaction can influence cancer progression. In tooth agenesis insufficient BarX1 activity can impair the formation of dental tissues resulting in missing teeth. Understanding these associations aids in developing targeted therapeutic interventions.

Target data

Transcription factor, which is involved in craniofacial development, in odontogenesis and in stomach organogenesis. May have a role in the differentiation of molars from incisors. Plays a role in suppressing endodermal Wnt activity (By similarity). Binds to a regulatory module of the NCAM promoter.
