Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Bassoon/BSN antibody [L124-59] - BSA and Azide Free (AB306594)
This data was developed using ab306593, the same antibody clone in a different buffer formulation. Immunohistochemical analysis of paraffin-embedded (A) HEK-293T cells transfected with a BSN expression vector containing a his tag. (B) HEK-293T cells transfected with empty vector containing a his tag stained for Bassoon/BSN using ab306593 at a 1/20000 dilution, followed by ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Positive staining on (A) HEK-293T cells transfected with a BSM expression vector containing a his tag. No staining on (B) HEK-293T cells transfected with empty vector containing a his tag. The section was incubated with ab306593 for 30 mins at room temperature, followed by anti-mouse IgG2b antibody (ab190482) for 8 mins during the LeicaDS9800 kit staining procedure. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond® Polymer Refine Detection). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Biological function summary
Bassoon plays an important role in the presynaptic active zone by facilitating efficient synaptic vesicle docking and priming. It acts as a scaffold protein within the presynaptic terminal forming part of a larger molecular complex that includes Piccolo and other cytomatrix proteins. These scaffolding arrangements help ensure precise neurotransmitter release affecting synaptic strength and plasticity. Its integration into these complexes highlights its significance in neuronal communication and functionality.
Pathways
Bassoon actively participates in neurotransmission pathways especially those related to synaptic vesicle cycle and neurotransmitter release. It collaborates with proteins like RIM and Munc13 in these processes stabilizing the apparatus required for synaptic vesicle fusion and exocytosis. Through these interactions Bassoon contributes to the regulation of neurotransmitter release and is essential for proper synaptic function.
