Supplementary information

Beta 1 Spectrin also known as SPTB is a large cytoskeletal protein with a mass of approximately 247 kDa. It mechanically contributes to the stabilization of the plasma membrane by forming a meshwork beneath it. This protein links the plasma membrane to the actin cytoskeleton. SPTB is abundantly expressed in erythrocytes and neurons where it plays important roles in maintaining cell shape and structural integrity.
Biological function summary

Beta 1 Spectrin is part of the spectrin-actin cytoskeleton complex and participates in cellular functions such as maintaining membrane integrity and flexibility. It anchors various proteins to the cytoskeleton and regulates membrane protein distribution. The interactions of beta 1 Spectrin within cells ensure proper localization of membrane channels and receptors critical for cellular signaling and transport activities.

Pathways

Beta 1 Spectrin functions in pathways connected to membrane cytoskeleton dynamics and cellular environmental responses. It closely relates to the ankyrin pathway where it interacts with ankyrin proteins to connect spectrin-actin network to plasma membrane proteins. Furthermore beta 1 Spectrin is involved in the maintenance and organization of lipid rafts which are specialized membrane microdomains enriched with specific lipids and proteins.

Beta 1 Spectrin mutations and defects associate with conditions like hereditary spherocytosis and spinocerebellar ataxia. In hereditary spherocytosis defects in beta 1 Spectrin lead to hemolytic anemia due to the altered erythrocyte membrane stability. This protein also interacts with ankyrin and band 3 proteins which are important in red blood cell membrane architecture and function. In spinocerebellar ataxia abnormalities in beta 1 Spectrin affect neuronal function contributing to progressive motor coordination issues.

Target data

Spectrin is the major constituent of the cytoskeletal network underlying the erythrocyte plasma membrane. It associates with band 4.1 and actin to form the cytoskeletal superstructure of the erythrocyte plasma membrane.
