Blocking and diluting buffer and concentration : 5% NFDM/TBST.

Performed under reducing conditions.

In Western blot, ab324049 was shown to bind specifically to Beta TRCP/HOS. Target of interest was observed at 60 kDa in wild-type HEK-293 cell lysates (lane 1) with no signal observed at this size in BTRC knockout cell line (lane 2) (knockout cell line ab266201 / knockout cell lysate ab257864).

To minimize protein degradation, cells were lysed immediately after harvest and then applied to a gel and transfer membrane for Western blotting as soon as possible. When lysates were stored at -80℃ prior to Western Blotting degradation products were observed.
