May be required for normal outer mitochondrial membrane dynamics (PubMed:15452144). Required for coatomer-mediated retrograde transport in certain cells (By similarity). May recruit other proteins to membranes with high curvature. May promote membrane fusion (PubMed:11604418). Involved in activation of caspase-dependent apoptosis by promoting BAX/BAK1 activation (PubMed:16227588). Isoform 1 acts proapoptotic in fibroblasts (By similarity). Involved in caspase-independent apoptosis during nutrition starvation and involved in the regulation of autophagy. Activates lipid kinase activity of PIK3C3 during autophagy probably by associating with the PI3K complex II (PI3KC3-C2) (PubMed:17891140). Associated with PI3KC3-C2 during autophagy may regulate the trafficking of ATG9A from the Golgi complex to the peripheral cytoplasm for the formation of autophagosomes by inducing Golgi membrane tubulation and fragmentation (PubMed:21068542). Involved in regulation of degradative endocytic trafficking and cytokinesis, probably in the context of PI3KC3-C2 (PubMed:20643123). Isoform 2 acts antiapoptotic in neuronal cells; involved in maintenance of mitochondrial morphology and promotes neuronal viability (By similarity).

Bif-1 also known as BAX-interacting factor-1 is a protein involved in the regulation of apoptosis and autophagy. It has an approximate mass of 23 kDa and is expressed in various tissues including the brain and immune cells. The protein is an important player in mitochondrial dynamics participating in membrane curvature required for mitochondrial fission.
This protein participates in mitochondrial fragmentation and membrane remodeling. Bif-1 is a component of the endophilin B1 complex which is involved in the regulation of apoptosis. It interacts directly with BAX a pro-apoptotic protein and enhances BAX translocation to mitochondria thereby facilitating the release of cytochrome c and subsequent cell death processes. It also influences autophagic processes by binding to Beclin1.
Bif-1 functions as an important link between the apoptosis and autophagy pathways. It facilitates the intrinsic apoptotic signaling pathway where BAX and Beclin1 play significant roles. In these pathways the protein controls cellular homeostasis and stress responses linking to other key regulators such as Bak and p53 which are involved in tumor suppression and DNA damage responses.
Bif-1 has connections to neurodegenerative diseases and cancer. Its dysregulation is associated with impaired autophagy and apoptosis contributing to the pathogenesis of Alzheimer's disease. In cancers altered expression of Bif-1 affects tumor growth and resistance to cell death interacting with proteins such as p53 and BAX which are pivotal in controlling cellular proliferation and survival.
