Formalin-fixed, paraffin embedded human prostate tissue stained for AKT1 (left) using ab272113 at 20 μg/mL, followed by Streptavidin-HRP at 10 μg/mL. Nuclear counterstain : hematoxylin. Antigen retrieval : Citrate, pH 6.2, pressure cooker.
Negative control (right) used pH9 antigen retrieval.

Immunohistochemical staining of AKT (phospho S473) in FFPE mouse prostate tissue. Tissue was fixed in 10% buffered formalin and subjected to heat‑mediated antigen retrieval using citrate buffer (pH 6.2) in a pressure cooker. The primary antibody (AKT pS473, biotin) was applied at 20 µg/mL for 1 hour at room temperature, followed by Streptavidin‑HRP (10 µg/mL) as the secondary detection reagent. Staining is predominantly nuclear with occasional cytoplasmic signal. Detection was performed using a red chromogenic precipitate, with hematoxylin providing a purple nuclear counterstain. Images shown at 40× magnification, with the stained sample on the left and the negative control on the right.
