Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Biotin Anti-Cytokeratin 17 antibody [EP1623] - Cytoskeleton Marker (AB195906)
IHC image of Cytokeratin 17 staining in a section of formalin-fixed paraffin-embedded normal human skin*.
The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6) in a Biocare Medical NxGen pressure cooker using retrieval settings of 110oC for 8 minutes. Non-specific protein-protein interactions were then blocked in TBS containing 0.025% (v/v) Triton X-100, 0.3M (w/v) glycine and 1% (w/v) BSA for 1h at room temperature. The section was then incubated overnight at +4°C with ab195906 at 1/100 dilution. DAB was used as the chromogen (ab103723), diluted 1/100 and incubated for 10min at room temperature. The section was counterstained with haematoxylin and mounted with DPX. The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

K17 often forms a complex with its partner keratin 6 enabling structural stability and resilience in epithelial cells.
