Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Biotin Anti-Glucagon antibody [EP3070] (AB314399)
Negative staining of glucagon in a section of formalin-fixed paraffin-embedded mouse normal spleen. Performed on a Leica BOND™ system. The section was pre-treated using heat mediated antigen retrieval with EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20mins, before blocking of endogenous biotin using ab64212. The section was then incubated with ab314399 at 0.06 µg /ml, for 15 mins at room temperature and detected using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset secondary-only control image is taken from an identical assay without primary antibody. For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
Glucagon plays a central role in maintaining glucose homeostasis. It increases blood glucose by promoting gluconeogenesis and glycogenolysis in the liver. Though glucagon mainly acts independently it exhibits significant interactions with other metabolic hormones such as insulin. This interaction helps balance blood sugar levels as glucagon and insulin work in opposition to ensure optimal blood glucose regulation.
Pathways
Glucagon primarily impacts the cAMP signaling pathway significantly increasing intracellular cAMP levels. This pathway initiates a complex series of downstream events including increased enzyme activity for gluconeogenesis and glycogen breakdown in the liver. Glucagon also cross-talks with insulin signaling pathways enabling the intertwined regulation of metabolism through these hormones and maintaining glucose balance.
