ELISA of human immunoglobulins shows ab224181 only reacted to human IgG3. No cross reactivity with Human IgG1, IgG2, IgG4, IgE, IgD, IgA, mouse IgG, rat IgG, or goat IgG. The plate was coated with 50 ng/well of different immunoglobulins. 200 ng/mL, 50 ng/mL, or 10 ng/mL of ab224181 was used as the primary antibody. An alkaline phosphatase conjugated anti-rabbit IgG was used as the secondary antibody.

IgG3 also known as gamma rat IgG3 and 1gG3 is an immunoglobulin G subclass involved in immune response. It has a molecular mass of approximately 150 kDa. This antibody is expressed in B cells and circulates throughout the bloodstream. It plays a mechanical role in immune defense by recognizing and binding to antigens assisting in pathogen neutralization and immune activation.
Biological function summary

IgG3 is part of a complex network within the immune system. It binds to native proteins on the surface of pathogens enhancing their recognition by phagocytes. IgG3 function includes efficient complement activation which is critical for the opsonization and elimination of antigens. Constant region of immunoglobulin heavy chains. Immunoglobulins, also known as antibodies, are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound immunoglobulins serve as receptors which, upon binding of a specific antigen, trigger the clonal expansion and differentiation of B lymphocytes into immunoglobulins-secreting plasma cells. Secreted immunoglobulins mediate the effector phase of humoral immunity, which results in the elimination of bound antigens (PubMed : 20176268, PubMed : 22158414). The antigen binding site is formed by the variable domain of one heavy chain, together with that of its associated light chain. Thus, each immunoglobulin has two antigen binding sites with remarkable affinity for a particular antigen. The variable domains are assembled by a process called V-(D)-J rearrangement and can then be subjected to somatic hypermutations which, after exposure to antigen and selection, allow affinity maturation for a particular antigen (PubMed : 17576170, PubMed : 20176268).
