Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Biotin Anti-IRF4 antibody [EP5699] (AB201482)
IHC image of IRF4 staining in a section of formalin-fixed paraffin-embedded human lymph node (Hodgkin's lymphoma)*, performed on a Leica Bond system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab201482 at 1/100 dilution for 15 mins at room temperature and detected using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
IRF4 helps in the activation and differentiation of immune cells. It forms complexes with other transcription factors like PU.1 enhancing its regulatory functions. IRF4 also contributes to the maturation of B and T lymphocytes impacting the adaptive immune system. It acts by modulating expression of cytokines and surface markers involved in immune response.
Pathways
IRF4 functions within the immune response and cell differentiation pathways. It interacts with pathways like NF-kB and MAPK linking it to important immune regulatory processes. IRF4 associates with proteins such as c-Rel in the NF-kB pathway influencing gene expression related to immune activation and survival.
