Activity summary

Macrophage Inflammatory Protein 1 gamma (MIP-1γ) also known as CCL9 functions as a chemokine involved in immune cell signaling. It weighs approximately 11 kDa. MIP-1γ attracts various immune cells to sites of inflammation within the body. The protein is expressed mainly in macrophages but you can also find it in other immune cells like dendritic cells. It plays a significant role in the body's response to inflammatory stimuli by mediating the recruitment of leukocytes.

Biological function summary

MIP-1γ contributes to immune response modulation by acting as a signaling molecule. It helps coordinate the movement and activation of immune cells such as monocytes and lymphocytes thereby modulating inflammation. Unlike some other chemokines MIP-1γ does not usually form a complex with proteins. Its primary role involves direct interactions with specific receptors on target cells that initiate intracellular signaling pathways leading to cell migration and activation.

Pathways

MIP-1γ engages in chemokine signaling and inflammatory response pathways. It intricately associates with the chemokine receptor CCR1 which is vital for mediating signal transduction processes essential for cell movement and immune surveillance. MIP-1γ also plays a role in the NF-kB signaling pathway important for transcription regulation and inflammatory response. This relationship highlights its involvement in orchestrating immune cell migration and the regulation of immune responses.

Associated diseases and disorders

MIP-1γ connects to various conditions including chronic inflammatory diseases and certain cancers. For example it has a link to rheumatoid arthritis where it contributes to the persistence of inflammation by recruiting immune cells to affected joints. It is also associated with some types of leukemia where its expression levels can influence disease progression. Proteins like interleukin-1β interact with MIP-1γ in these conditions as they both participate in inflammatory responses that exacerbate disease states.

    To detect Murine MIP-1γ by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 μg/ml.

    When used in conjunction with compatible secondary reagents, the detection limit for recombinant Murine MIP-1γ is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

    Lane 1: Marker

    Lanes 2-12: 250, 125, 62.5, 31.25, 15.625, 7.8, 3.9, 1.95, 0.975, 0.4875 and 0.24 ng recombinant mouse Macrophage Inflammatory Protein 1 gamma, respectively.

    Mouse MIP-1γ was detected by sandwich ELISA (using 100μl/well) using a concentration of 0.25-1.0 μg/ml of ab271205.

    This antibody in conjunction with an appropriate capture antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant mMIP-1γ protein.
