Target data

Function

Growth factor of the TGF-beta superfamily that plays important role in various biological processes, including embryogenesis, hematopoiesis, neurogenesis and skeletal morphogenesis (PubMed:31208997). Initiates the canonical BMP signaling cascade by associating with type I receptor ACVR1 and type II receptor ACVR2A (PubMed:12667445, PubMed:9748228). Once all three components are bound together in a complex at the cell surface, ACVR2A phosphorylates and activates ACVR1. In turn, ACVR1 propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target genes (PubMed:12478285). For specific functions such as growth cone collapse in developing spinal neurons and chemotaxis of monocytes, uses also BMPR2 as type II receptor (PubMed:31208997). Can also signal through non-canonical pathways such as P38 MAP kinase signaling cascade that promotes brown adipocyte differentiation through activation of target genes, including members of the SOX family of transcription factors (PubMed:27923061). Promotes the expression of HAMP, this is repressed by its interaction with ERFE (PubMed:30097509).

    Immunoprecipitation - Anti-BMP7 antibody [EPR23143-31] (ab269586)

BMP7 was immunoprecipitated from 0.35 mg MCF7 (Human breast adenocarcinoma epithelial cell) whole cell lysate 10μg with ab269586 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab269586 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (VeriBlot for IP Detection Reagent (HRP) ab131366) was used at 1/1000 dilution.

    Lane 1: MCF7 whole cell lysate 10μg

Lane 2: ab269586 IP in MCF7 whole cell lysate

Lane 3: Rabbit monoclonal IgG (Rabbit IgG, monoclonal [EPR25A] - Isotype Control ab172730) instead of ab269586 in MCF7 whole cell lysate

Blocking and dilution buffer and concentration: 5% NFDM/TBST.

    Exposure time: 3 seconds.

    All lanes: Immunoprecipitation - Anti-BMP7 antibody [EPR23143-31] (ab269586)

    Predicted band size: 49 kDa

    Western blot - Anti-BMP7 antibody [EPR23143-31] (ab269586)

Blocking and diluting buffer and concentration: 5% NFDM/TBST

The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID:16419056, 12792780,11807825)

    Negative control: MDA-MB-231 (PMID: 16419056)

This blot was developed using a higher sensitivity ECL substrate. Exposure time: 3 minutes

    All lanes: Western blot - Anti-BMP7 antibody [EPR23143-31] (ab269586) at 1/1000 dilution

Lane 1: MCF7 (human breast adenocarcinoma epithelial cell) whole cell lysate at 20 µg

Lane 2: MDA-MB-231 (human breast adenocarcinoma epithelial cell) whole cell lysate at 20 µg

    Secondary

All lanes: Western blot - Goat Anti-Rabbit IgG H&L (HRP) (Goat Anti-Rabbit IgG H&L (HRP) ab97051) at 1/100000 dilution

    Predicted band size: 49 kDa

    Observed band size: 49 kDa
