Supplementary information

BTN3A1 also known as CD277 is an immunoregulatory protein with a mass of approximately 57 kDa. It belongs to the butyrophilin family part of the immunoglobulin superfamily. BTN3A1 is expressed in a variety of tissues including the immune system where it is present on the surface of immune cells such as T cells and natural killer cells. The protein plays a role in the immune cell activation and modulation.
Biological function summary

BTN3A1 functions as a co-stimulatory molecule in immune responses. It forms part of a receptor complex that modulates T cell activation and cytokine production. BTN3A1 is essential for the recognition and response to phosphoantigens which are small phosphorylated metabolites. This function allows BTN3A1 to regulate immune responses effectively and shape the body's defense against infections.

Pathways

BTN3A1 plays a critical role in the regulation of the γΔ T cell activation pathway which is important for immune responses against a variety of pathogens. BTN3A1 interacts with other proteins such as BTN3A2 and B7-H3 facilitating the activation and function of γΔ T cells. These interactions influence the immune system's ability to respond appropriately to infections or malignancies.

BTN3A1 has been implicated in autoimmune diseases and cancer. Alterations in BTN3A1 expression or function can contribute to autoimmune disorders due to dysregulated immune responses. Additionally BTN3A1 is studied in the context of cancer where its interactions with proteins like BTN3A3 may influence tumor immune escape mechanisms. Understanding BTN3A1's role in these conditions provides insights into potential therapeutic targets for managing autoimmune diseases and cancer.

Target data

Plays a role in T-cell activation and in the adaptive immune response. Regulates the proliferation of activated T-cells. Regulates the release of cytokines and IFNG by activated T-cells. Mediates the response of T-cells toward infected and transformed cells that are characterized by high levels of phosphorylated metabolites, such as isopentenyl pyrophosphate.
