Bub3 also known as budding uninhibited by benzimidazoles-3 is an important component of the spindle checkpoint machinery. Weighing approximately 35 kDa this protein is expressed ubiquitously in eukaryotic cells. Bub3 interacts with other spindle checkpoint proteins and takes part in chromosome segregation during mitosis. It primarily functions in the nucleus during the cell cycle ensuring proper attachment of chromosomes to the mitotic spindle which is important for accurate cell division.
Biological function summary

Bub3 participates in the mitotic checkpoint complex an essential mediator of cell cycle arrest in response to improper spindle attachment. This complex includes Bub1 BubR1 and Mad3 where Bub3 acts as an adapter facilitating interactions within the complex. It helps maintain chromosomal stability by preventing premature anaphase onset therefore ensuring that damaged or unaligned chromosomes do not progress through cell division. By doing so Bub3 safeguards against aneuploidy and genomic instability.

Pathways

Bub3 is linked to the spindle assembly checkpoint (SAC) and the cell cycle regulation pathway. In SAC Bub3 collaborates closely with proteins such as Bub1 and Mad1 to inhibit the anaphase-promoting complex/cyclosome (APC/C) until all chromosomes are properly aligned. The interplay between Bub3 and BubR1 is important for delaying the chief exit phases of mitosis ensuring that all cellular conditions are favorable for division. These interactions prevent errors that might cause irreparable damage or cell death.

Bub3 has associations with cancer and some neurodegenerative diseases. Its dysregulation can lead to aneuploidy a hallmark of many cancers through faulty chromosome segregation. In several cancer types altered expression of Bub3 and its partners such as Bub1 and Mad2 has been documented. Similarly emerging studies suggest links between Bub3 malfunction and neurodegeneration where alterations in cell division and genomic stability may contribute to disease progression.

Has a dual function in spindle-assembly checkpoint signaling and in promoting the establishment of correct kinetochore-microtubule (K-MT) attachments. Promotes the formation of stable end-on bipolar attachments. Necessary for kinetochore localization of BUB1. Regulates chromosome segregation during oocyte meiosis. The BUB1/BUB3 complex plays a role in the inhibition of anaphase-promoting complex or cyclosome (APC/C) when spindle-assembly checkpoint is activated and inhibits the ubiquitin ligase activity of APC/C by phosphorylating its activator CDC20. This complex can also phosphorylate MAD1L1.
