Supplementary information

C-IAP1 and c-IAP2 also known as cellular inhibitors of apoptosis proteins 1 and 2 are important regulators of apoptosis and inflammation. c-IAP1 has a molecular weight of approximately 70 kDa while c-IAP2 is around 74 kDa. Both proteins are expressed in many tissues including the heart liver and immune cells. They function as E3 ubiquitin ligases where they target specific proteins for ubiquitination leading to proteasomal degradation. This activity plays a significant role in controlling cell survival and inflammation by modulating the levels of key signaling proteins.
Biological function summary

C-IAP1 and c-IAP2 contribute to cell survival by inhibiting apoptosis. They are part of a complex that interacts with other proteins and signaling molecules to regulate the apoptotic machinery. c-IAP1 and c-IAP2 interact with members of the tumor necrosis factor (TNF) receptor-associated factors (TRAFs) family which helps suppress programmed cell death and support cell survival. By doing so they maintain cellular homeostasis and balance between life and death signals in various physiological contexts.

Pathways

C-IAP1 and c-IAP2 are key components in the NF-kB and MAPK signaling pathways. These pathways are important for immune response inflammation and cell survival. c-IAPs interact with TNF receptor 1 (TNFR1) and receptor-interacting protein kinases (RIPKs) modulating downstream effects that activate NF-kB which transcribes genes involved in immune and survival responses. This connectivity highlights the involvement of c-IAP1 and c-IAP2 in pivotal signaling frameworks that coordinate cellular responses under stress and external stimuli.

C-IAP1 and c-IAP2 have significant roles in cancer and chronic inflammatory diseases. They are often overexpressed in various cancers including breast and lung cancer where they contribute to tumorigenesis by preventing cancer cell apoptosis and promoting cell survival. In the context of chronic inflammation these proteins maintain aberrant cell survival contributing to diseases such as rheumatoid arthritis. Connections with proteins like IκB kinase (IKK) highlight their impact on the perpetuation and progression of inflammatory conditions through their modulation of NF-kB signaling.

Target data

Multi-functional protein which regulates not only caspases and apoptosis, but also modulates inflammatory signaling and immunity, mitogenic kinase signaling, and cell proliferation, as well as cell invasion and metastasis. Acts as an E3 ubiquitin-protein ligase regulating NF-kappa-B signaling and regulates both canonical and non-canonical NF-kappa-B signaling by acting in opposite directions : acts as a positive regulator of the canonical pathway and suppresses constitutive activation of non-canonical NF-kappa-B signaling. The target proteins for its E3 ubiquitin-protein ligase activity include : RIPK1, RIPK2, RIPK3, RIPK4, CASP3, CASP7, CASP8, TRAF2, DIABLO/SMAC, MAP3K14/NIK, MAP3K5/ASK1, IKBKG/NEMO, IKBKE and MXD1/MAD1. Can also function as an E3 ubiquitin-protein ligase of the NEDD8 conjugation pathway, targeting effector caspases for neddylation and inactivation. Acts as an important regulator of innate immune signaling via regulation of Toll-like receptors (TLRs), Nodlike receptors (NLRs) and RIG-I like receptors (RLRs), collectively referred to as pattern recognition receptors (PRRs). Protects cells from spontaneous formation of the ripoptosome, a large multi-protein complex that has the capability to kill cancer cells in a caspase-dependent and caspase-independent manner. Suppresses ripoptosome formation by ubiquitinating RIPK1 and CASP8. Can stimulate the transcriptional activity of E2F1. Plays a role in the modulation of the cell cycle.
