C10orf63 also known as TMEM119 is a protein that plays a critical role in cellular processes. It has a molecular weight of approximately 54 kDa and is predominantly expressed in microglial cells within the central nervous system. TMEM119 localizes to the plasma membrane contributing to the regulation of specific signaling pathways.
TMEM119 serves to regulate immune responses in the brain participating as part of a larger complex involving other membrane proteins. It is integral in maintaining microglial identity and function. TMEM119 influences processes such as cell activation migration and phagocytosis. By doing so it contributes to the homeostasis and defense mechanisms in the neural environment.
TMEM119 is involved in the TLR4 signaling pathway which is essential for the innate immune responses. It interacts with proteins such as MyD88 which transduces downstream inflammatory signaling cascades. TMEM119's role in this pathway ensures precise control over inflammatory responses and cellular activation which are critically important in the brain's microenvironment.
TMEM119 has a notable connection to neuroinflammation and neurodegenerative diseases particularly Alzheimer's disease and multiple sclerosis. In Alzheimer's disease TMEM119 interaction with proteins like TREM2 suggests a role in modulating inflammatory responses and amyloid-beta phagocytosis. Similarly in multiple sclerosis altered TMEM119 expression can influence microglial activity and contribute to disease progression revealing its importance in pathological conditions.
