Supplementary information

C14orf166 also known as TIPH2 is a protein encoded by the C14orf166 gene. This protein has a molecular mass of approximately 38 kDa. It plays a role in RNA metabolism particularly in the export of messenger RNA (mRNA) from the nucleus to the cytoplasm. Expression of C14orf166 occurs in various tissues with higher levels often found in actively dividing and metabolically active cells.
Biological function summary

C14orf166 integrates into the transcriptional regulation process by forming part of a complex involved in RNA processing. It interacts with components of the transcription-export (TREX) complex which is essential for coupling mRNA processing with export. This integration allows C14orf166 to assist in maintaining efficient mRNA transport and translation important for normal cellular function and stress response.

Pathways

C14orf166 significantly influences gene expression regulation and participates in the mRNA processing pathways. It interacts with proteins such as Aly/REF and UAP56 within the TREX complex importantly impacting the efficiency of mRNA export. By securing efficient mRNA transport C14orf166 supports critical biological pathways like the unfolded protein response and cellular proliferation.

Aberrant expression or function of C14orf166 has connections to cancer particularly breast cancer. Dysregulation in its function can lead to improper mRNA export contributing to tumorigenesis. Furthermore C14orf166 shows relations to viral infections; for example it intersects with pathways involving Nucleophosmin (NPM1) which are exploited by viruses such as influenza. This makes C14orf166 a potential target for therapeutic strategies aiming to manage these conditions.

Target data

Accessory subunit of the tRNA-splicing ligase complex that acts by directly joining spliced tRNA halves to mature-sized tRNAs by incorporating the precursor-derived splice junction phosphate into the mature tRNA as a canonical 3',5'-phosphodiester (PubMed : 21311021, PubMed : 24870230). RNA-binding protein involved in modulation of mRNA transcription by Polymerase II (PubMed : 16950395). Could also play a role in RNA transport (PubMed : 24608264).. (Microbial infection) In case of infection by influenza virus A (IVA), is involved in viral replication (PubMed : 21900157).
