Activation of C5 by a C5 convertase initiates the spontaneous assembly of the late complement components, C5-C9, into the membrane attack complex. C5b has a transient binding site for C6. The C5b-C6 complex is the foundation upon which the lytic complex is assembled. C5a anaphylatoxin. Derived from proteolytic degradation of complement C5, C5a anaphylatoxin is a mediator of local inflammatory process. Binding to the receptor C5AR1 induces a variety of responses including intracellular calcium release, contraction of smooth muscle, increased vascular permeability, and histamine release from mast cells and basophilic leukocytes (PubMed:8182049). C5a is also a potent chemokine which stimulates the locomotion of polymorphonuclear leukocytes and directs their migration toward sites of inflammation.

The C5a protein also known as anaphylatoxin C5a is a small potent molecule weighing approximately 11 kDa. It originates from the larger complement component C5 during complement system activation. The C5a des Arg is a slightly modified version produced when enzymes cleave the C-terminal arginine from C5a. Both forms express on cell surfaces and recognize specific receptors on target cells particularly in immune and inflammatory responses.
This protein controls complex interactions within the immune system specifically as part of the cleavage product of C5 a component of the complement system. C5a binds to receptors such as C5aR and C5L2 which appear in high numbers on various immune cells like macrophages neutrophils and endothelial cells. This binding triggers multiple intracellular pathways mediating inflammatory processes and immune cell activation.
The complement system involves the activation sequences where C5a plays a pivotal role. It participates in the activation of the classic and the alternative complement pathways contributing to the opsonization and lysis of pathogens. C5a works closely with proteins such as C3a and C9 orchestrating the body's defense mechanisms against infections while facilitating communication between the innate and adaptive immune systems.
C5a plays a notable role in inflammatory diseases like rheumatoid arthritis and sepsis. Its overactivation relates to severe inflammatory responses and tissue damage.
