C8G also known as complement component 8 gamma plays a significant role in the immune system's function. This protein has a mass of approximately 22 kDa and forms a part of the complement component 8 complex which acts as a critical player in the formation of the Membrane Attack Complex (MAC). Researchers have found C8G in various tissues with notable expression in the liver. The protein is generally released into the bloodstream as part of its involvement in immune responses.
Biological function summary

Complement component 8 gamma participates in puncturing pathogen cell membranes. It functions within the Membrane Attack Complex a structure important for antimicrobial activity. C8G partners with complement components C8 alpha and C8 beta to form the complete C8 complex which then associates with other complement proteins to facilitate membrane penetration. This activity highlights its role in protecting the host from infectious agents by disrupting pathogen integrity.

Pathways

C8G contributes critically to the complement cascade particularly within the terminal pathway. This pathway involves other complement proteins like C5b C6 and C7 coordinating to assemble the MAC which serves as a defense mechanism against microbial invasion. Consistently C8G shares functional relationships with these proteins reinforcing its participation in immune mediators.

C8G dysfunction associates with conditions like Complement Component 8 Deficiency which results in increased susceptibility to Neisseria infections. Target data

Component of the membrane attack complex (MAC), a multiprotein complex activated by the complement cascade, which inserts into a target cell membrane and forms a pore, leading to target cell membrane rupture and cell lysis (PubMed : 26841837, PubMed : 27052168, PubMed : 30552328). The MAC is initiated by proteolytic cleavage of C5 into complement C5b in response to the classical, alternative, lectin and GZMK complement pathways (PubMed : 30552328, PubMed : 39914456, PubMed : 39814882). The complement pathways consist in a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed : 30552328). C8G, together with C8A and C8B, inserts into the target membrane, but does not form pores by itself (PubMed : 30552328). During MAC assembly, associates with C5b, C6 and C7 to form the C5b8 intermediate complex that inserts into the target membrane and traverses the bilayer increasing membrane rigidity (PubMed : 30552328, PubMed : 6833260).
