C8 is a constituent of the membrane attack complex. C8 binds to the C5B-7 complex, forming the C5B-8 complex. C5-B8 binds C9 and acts as a catalyst in the polymerization of C9. The gamma subunit seems to be able to bind retinol.

C8G also known as complement component 8 gamma plays a significant role in the immune system's function. This protein has a mass of approximately 22 kDa and forms a part of the complement component 8 complex which acts as a critical player in the formation of the Membrane Attack Complex (MAC). Researchers have found C8G in various tissues with notable expression in the liver. The protein is generally released into the bloodstream as part of its involvement in immune responses.
Complement component 8 gamma participates in puncturing pathogen cell membranes. It functions within the Membrane Attack Complex a structure important for antimicrobial activity. C8G partners with complement components C8 alpha and C8 beta to form the complete C8 complex which then associates with other complement proteins to facilitate membrane penetration. This activity highlights its role in protecting the host from infectious agents by disrupting pathogen integrity.
C8G contributes critically to the complement cascade particularly within the terminal pathway. This pathway involves other complement proteins like C5b C6 and C7 coordinating to assemble the MAC which serves as a defense mechanism against microbial invasion. Consistently C8G shares functional relationships with these proteins reinforcing its participation in immune mediators.
C8G dysfunction associates with conditions like Complement Component 8 Deficiency which results in increased susceptibility to Neisseria infections. Moreover alterations in C8G can link to systemic lupus erythematosus an autoimmune disease where complement activation may become dysregulated. C8G's interactions with C8 alpha and C8 beta components remain important in these pathophysiological contexts potentially serving as targets for therapeutic interventions aimed at modulating complement activity.

An Alexa Fluor®555-conjugated goat anti-rabbit IgG was used as a secondary antibody at 1/200 dilution. Nuclei were counterstained with DAPI (blue).
