Supplementary information

C9ORF46 also known as PLGRKT is a protein encoded by the C9ORF46 gene. It has a molecular mass of approximately 28 kDa. Scientists have detected its expression in various tissues with particular enrichment in neurons. This protein consists of a unique domain structure including a combination of leucine-rich domains. As an intracellular component PLGRKT may interact with other proteins or membranes within the cell.
Biological function summary

C9ORF46/PLGRKT plays a role in cellular processes that involve the regulation of fibrinolysis. It does not act in isolation and is known to be part of complexes that modulate other proteins' activity. The protein serves as an important mediator in tissue remodeling and wound healing. By interacting with enzymes such as plasminogen it influences the actions on the extracellular matrix components affecting cell migration and tissue repair mechanisms.

Pathways

C9ORF46 influences the plasminogen activation system. This protein's role in the fibrinolytic pathway is key for promoting the breakdown of fibrin a protein involved in blood clotting. Additionally C9ORF46 is related to the urokinase-type plasminogen activator (uPA) through this pathway. Together they coordinate in controlling the conversion of plasminogen to its active form plasmin which degrades fibrin and other matrix proteins.

C9ORF46/PLGRKT is linked to the regulation of processes relevant to thrombotic conditions. Abnormal function or expression of this protein can contribute to disorders of fibrinolysis such as thrombosis. Moreover its interaction with other proteins like the aforementioned plasminogen can lead to impaired wound healing. Understanding the precise relationship between C9ORF46 and these conditions could reveal potential therapeutic targets for treatment.

Target data

Receptor for plasminogen. Regulates urokinase plasminogen activator-dependent and stimulates tissue-type plasminogen activator-dependent cell surface plasminogen activation. Proposed to be part of a local catecholaminergic cell plasminogen activation system that regulates neuroendocrine prohormone processing. Involved in regulation of inflammatory response; regulates monocyte chemotactic migration and matrix metalloproteinase activation, such as of MMP2 and MMP9.
