The alpha-2/delta subunit of voltage-dependent calcium channels regulates calcium current density and activation/inactivation kinetics of the calcium channel. Acts as a regulatory subunit for P/Q-type calcium channel (CACNA1A), N-type (CACNA1B), L-type (CACNA1C OR CACNA1D) and possibly T-type (CACNA1G) (PubMed:15111129, PubMed:23339110). Overexpression induces apoptosis.

CACNA2D2 also known as calcium channel voltage-dependent alpha 2/delta subunit 2 is a protein involved in the function of voltage-gated calcium channels. This protein acts as an auxiliary subunit and plays a role in the regulation of the channel's currents. CACNA2D2 has a mass of approximately 129 kDa and is typically expressed in various tissues including the brain and heart. It shows significant involvement in neurons playing a role in neurotransmitter release due to its modulation of calcium entry into cells.
CASNA2D2 is important for the function of nervous and cardiac systems. It forms part of a larger protein complex that includes alpha-1 beta and gamma subunits which all work together to influence calcium influx in response to membrane depolarization. This calcium entry is important for processes such as synaptic transmission and muscle contraction. CACNA2D2's modulation of calcium channels hence affects the excitability of neurons and the contractility of cardiac muscle cells impacting both neuronal signaling and heart function.
The presence of CACNA2D2 influences the calcium signaling pathway and the cardiac conduction pathway. This protein modifies cellular processes by regulating the calcium ion concentration inside cells which can trigger various signaling cascades. It interacts closely with other proteins in these pathways like the L-type calcium channels which mediate significant increases in intracellular calcium levels. Such interactions further highlight its role in normal physiological responses to electrical stimuli in cells.
Alterations in CACNA2D2 expression or function can lead to epilepsy and cardiac arrhythmias. Changes in this protein's behavior can disrupt normal calcium homeostasis leading to altered neuronal firing patterns and impaired cardiac rhythm. Its relationship with epilepsy highlights its role as a potential target for anticonvulsant drugs aiming to stabilize neuronal firing. Similarly it connects with other proteins like CACNA1C in the context of cardiac diseases linking these proteins in the pathological manifestation of arrhythmias.
