Lane 2 : ACNA2D2 knockout HAP1 cell lysate (20 μg)

Lanes 1 - 4 : erged signal (red and green). Green - ab173293 observed at 160 kDa. Red - loading control, ab8245, observed at 37 kDa.

ab173293 was shown to specifically react with CACNA2D2 when CACNA2D2 knockout samples were used. Wild-type and CACNA2D2 knockout samples were subjected to SDS-PAGE. ab173293 and ab8245 (loading control to GAPDH) were diluted at 1/5000 and 1/10000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

CACNA2D2 also known as calcium channel voltage-dependent alpha 2/delta subunit 2 is a protein involved in the function of voltage-gated calcium channels. This protein acts as an auxiliary subunit and plays a role in the regulation of the channel's currents. CACNA2D2 has a mass of approximately 129 kDa and is typically expressed in various tissues including the brain and heart. Acts as a regulatory subunit for P/Q-type calcium channel (CACNA1A), N-type (CACNA1B), L-type (CACNA1C OR CACNA1D) and possibly T-type (CACNA1G) (PubMed : 15111129, PubMed : 23339110).
