Immunohistochemistry was performed on normal biopsies of deparaffinized Human skeletal muscle tissue. To expose target proteins heat induced antigen retrieval was performed using 10mM sodium citrate (pH6.0) buffer microwaved for 8-15 minutes. Following antigen retrieval tissues were blocked in 3% BSA-PBS for 30 minutes at room temperature. Tissues were then probed at a dilution of 1 : 20 with a rabbit polyclonal antibody recognizing PMCA3 ATPase ab3530 or without primary antibody (negative control) overnight at 4°C in a humidified chamber. Tissues were washed extensively with PBST and endogenous peroxidase activity was quenched with a peroxidase suppressor. Detection was performed using a biotin-conjugated secondary antibody and SA-HRP followed by colorimetric detection using DAB. Tissues were counterstained with hematoxylin and prepped for mounting.

The Calcium Pump PMCA3 ATPase also known as Plasma Membrane Calcium-Transporting ATPase 3 plays an important role in calcium homeostasis. This enzyme weighing approximately 134 kDa actively transports calcium ions out of cells to maintain low intracellular calcium levels. PMCA3 is mainly expressed in the brain and other neural tissues where it helps regulate neuronal signaling. The pump uses ATP to move calcium against its concentration gradient from the cytosol to the extracellular space a critical function for cellular activities.
Biological function summary

PMCA3 controls the concentration of cytosolic calcium ions which influences many cellular processes such as muscle contraction neurotransmitter release and gene expression. PMCA3 often functions independently although it can sometimes exist in complexes with other proteins influencing its regulation and activity. The regulation of neuronal calcium levels through PMCA3 is essential for proper synaptic function and plasticity making it a point of interest in neurophysiological research.

Pathways

PMCA3 plays a role in the calcium signaling pathway affecting signal transduction processes across various cell types.
