Pore-forming subunit of gustatory voltage-gated ion channels required for sensory perception of sweet, bitter and umami tastes. With CALHM1 forms a fast-activating voltage-gated ATP-release channel in type II taste bud cells, ATP acting as a neurotransmitter to activate afferent neural gustatory pathways. Acts both as a voltage-gated and calcium-activated ion channel: mediates neuronal excitability in response to membrane depolarization and low extracellular Ca(2+) concentration. Has poor ion selectivity and forms a wide pore (around 14 Angstroms) that mediates permeation of small ions including Ca(2+), Na(+), K(+) and Cl(-), as well as larger ions such as ATP(4-).

CALHM3 also known as calcium homeostasis modulator 3 is a voltage-gated ion channel that plays a critical role in cellular calcium signaling. It has a molecular mass of approximately 47 kDa. This protein forms a channel that facilitates the influx and efflux of calcium ions across the plasma membrane influencing membrane potential and cellular excitability. CALHM3 is predominantly expressed in taste bud cells cochlear cells and neurons. Its function is essential for processes that require ion homeostasis within these cells.
CALHM3 participates in taste perception by working with other proteins to form a pore complex that allows ATP release. This complex is important for signal transmission in taste receptor cells. The ability of CALHM3 channels to release ATP on taste bud stimulation is important for the communication of taste signals to sensory neurons. Through this interaction CALHM3 significantly influences the sensory response and perception of different tastes.
CALHM3 is part of the taste signaling pathway interacting with CALHM1 and other subunits to regulate gustatory signals. This pathway plays an important role in how organisms interpret various taste stimuli. Furthermore CALHM3's involvement in calcium signaling pathways connects it with other proteins such as P2X receptors which also mediate ATP signaling. These interactions demonstrate the protein's integration and contribution to essential sensory processes in the body.
Mutations in CALHM3 have links to taste disorders impacting taste perception and sensitivity known as ageusia or dysgeusia. Additionally its role in calcium signaling suggests potential connections to neurological conditions involving altered communication in neural circuits. Alterations in the CALHM3 structure or function can affect its interaction with CALHM1 impacting physiological processes tied to taste and potential neurodegenerative conditions.
