Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Calpain 10 antibody (AB28226)

IHC image of ab28226 staining in normal human pancreas formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab28226, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

ICC/IF image of ab28226 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab28226, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Calpain 10 also known as CAPN10 is an intracellular cysteine protease enzyme belonging to the calpain family. It has a molecular mass of approximately 75 kDa and shows expression in various tissues including the pancreas skeletal muscle and liver. Calpain 10 plays a mechanical role in modulating cellular processes by cleaving target substrates involved in cellular functions such as signal transduction differentiation and apoptosis. This enzyme requires calcium ions for its activation.
Biological function summary

Calpain 10 contributes to the regulation of intracellular signaling pathways and cellular homeostasis. It plays a role in cytoskeletal remodeling extricating proteins from multiprotein complexes and influencing glucose metabolism. Compared to other calpains Calpain 10 differs structurally as it lacks the typical EF-hand calcium-binding domains. This characteristic suggests its unique mechanism in modulating physiological processes within cells.

Pathways

Calpain 10 interacts with stress-related signaling and metabolic pathways. Particularly it functions within the insulin signaling pathway essential for glucose uptake and metabolism. Calpain 10’s activity affects the insulin receptor substrate (IRS) and is related to the phosphatidylinositol 3-kinase (PI3K) pathway which is important for cellular metabolism and survival. These pathways highlight the relationship between Calpain 10 and metabolic control.
