Supplementary information

Calpain 6 also known as CAPN6 is a protease without proteolytic activity and belongs to the calpain family. This protein weighs around 75 kDa and expresses mainly in the placenta embryonic tissues and a variety of tumor cells. Unlike other calpains CAPN6 does not have a conserved catalytic triad and acts as a scaffold rather than an proteolytic enzyme. It participates in cytoskeletal remodeling due to its strong interactions with microtubules.
Biological function summary

CAPN6 is involved in cell proliferation migration and growth regulation processes. The protein associates with microtubules contributing to the regulation of cellular mechanics and morphology. It interacts with other cellular components to support functions in tissue development and is important in maintaining cytoskeleton dynamics. CAPN6's engagement in cellular processes indicates its potential role as a cellular signaling mediator.

Pathways

CAPN6 is significantly involved in the Wnt signaling and cytoskeletal remodeling pathways. These pathways are essential for cellular growth and differentiation where CAPN6 modulates cytoskeletal rearrangements and influences these biological processes. In the Wnt pathway CAPN6 interacts with proteins like Dishevelled and influences the cellular structure and signaling outcomes involved in the pathway’s regulation.

CAPN6 is associated with various types of cancer and fibrosis processes. The aberrant expression of CAPN6 can lead to dysregulation in cell proliferation and mobility which connects directly to cancer progression especially in embryonic tumors and other malignancies. In cancer contexts CAPN6 associates with proteins such as β-catenin amplifying oncogenic signals through Wnt signaling making it a potential target for therapeutic interventions.

Target data

Microtubule-stabilizing protein that may be involved in the regulation of microtubule dynamics and cytoskeletal organization. May act as a regulator of RAC1 activity through interaction with ARHGEF2 to control lamellipodial formation and cell mobility. Does not seem to have protease activity as it has lost the active site residues (By similarity).
