Thin filament-associated protein that is implicated in the regulation and modulation of smooth muscle contraction. It is capable of binding to actin, calmodulin and tropomyosin. The interaction of calponin with actin inhibits the actomyosin Mg-ATPase activity.

Calponin 2 also known as CNN2 or basic calponin is an actin-binding protein with a molecular weight of approximately 33 kDa. It binds to thin filaments in a calcium-dependent manner stabilizing these structures within the cell. Calponin 2 predominantly expresses in smooth muscle tissues and various non-muscle cells where it influences cellular tension and mechanical processes such as contraction and motility. Its ability to interact with actin makes it important for modulating the dynamic behavior of the cytoskeleton.
Calponin 2 plays a role in regulating smooth muscle contraction and modulating cellular motility. In smooth muscle cells it is not firm component of a larger regulatory complex or tightly bounded to a specific protein partner but acts individually to mediate these biological actions. It also participates in non-muscle cell activities where it assists in stabilizing actin structures. Such activity is important for the maintenance of cellular architecture and function across various cell types.
Calponin 2 is intimately connected to the regulation of the RhoA/Rho kinase pathway and the actin cytoskeleton remodeling processes. This pathway is important for cellular motility adhesion and contraction as it involves interplay with actin. Calponin 2 interacts with proteins like tropomyosin and myosin linking its mechanical effects on actin filaments to broader cellular functions. Together these interactions contribute to the precise modulation of cellular behaviors essential for tissue formation and repair.
Calponin 2 associates with conditions such as asthma and vascular diseases. In asthma altered expression or function of Calponin 2 within smooth muscle cells can affect bronchial contractions leading to symptoms exacerbation. Similarly in vascular diseases defects in Calponin 2 function could impair smooth muscle contractility affecting blood vessel function. Its expression and interaction with myosin and tropomyosin in these disorders highlight its importance in maintaining smooth muscle cell functionality and contractile health.
