Calsequestrin is a high-capacity, moderate affinity, calcium-binding protein and thus acts as an internal calcium store in muscle (PubMed:28895244). Calcium ions are bound by clusters of acidic residues at the protein surface, often at the interface between subunits. Can bind around 80 Ca(2+) ions (PubMed:28895244). Regulates the release of lumenal Ca(2+) via the calcium release channel RYR1; this plays an important role in triggering muscle contraction. Negatively regulates store-operated Ca(2+) entry (SOCE) activity (PubMed:27185316).

Intracellular flow cytometric analysis of 2% paraformaldehyde fixed HeLa cells labeling Calsequestrin 1with ab185220 at 1/240 dilution (red) compared with a rabbit monoclonal IgG control (green). Goat anti rabbit IgG (FITC) at 1/150 dilution was used as the secondary antibody.
Blocking and diluting buffer and concentrartion: 5% NFDM/TBST.
Exposure time: Lane 1: 30 seconds; Lane 2: 180 seconds.
CASQ1 is the major isoform in adult fast-twitch skeletal muscles, while CASQ2 is the only isoform in cardiac muscle and a minor isoform in adult slow-twitch skeletal muscle (PMID: 33288873).
