Lane 2 : CAMK1D knockout HAP1 cell lysate (20 μg)

Lanes 1 - 4 : Merged signal (red and green). Green - ab172618 observed at 43 kDa. Red - loading control, ab18058, observed at 124 kDa.
ab172618 was shown to specifically react with CAMK1D when CAMK1D knockout samples were used. Wild-type and CAMK1D knockout samples were subjected to SDS-PAGE. ab172618 and ab18058 (loading control to Vinculin) were both diluted 1/10,000 and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

Western blot : Rabbit Monoclonal[EPR3536(2)] to CAMK1D ab172618 staining at 1/10000 dilution, shown in green; Mouse anti-CANX (ab238078) loading control staining at 1/20,000 dilution, shown in magenta. A band was observed at 42 kDa in Wild-type A549 cell lysates with no signal observed at this size in CAMK1D knockout A549 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3pc Milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW & Goat anti-Mouse 680RD at 1/20,000 dilution.

Biological function summary
CAMK1D participates in neuronal development and immune responses. It plays an important role in the regulation of gene expression and cytoskeletal rearrangement. CAMK1D does not typically form complexes but it interacts with other proteins to perform its functions. Its activity is regulated by the binding of calmodulin in response to increased intracellular calcium levels which activates its kinase activity leading to phosphorylation of target proteins.
Pathways
CAMK1D actively integrates into the calcium signaling pathway as well as the MAPK signaling pathway. Through these pathways it interacts with several proteins like CREB and acts to mediate the effects of calcium on cellular activities. Its role in calcium signaling links it to various responses such as cellular growth apoptosis and differentiation. CAMK1D shares functional interactions with other kinases in these pathways aiding in signal transduction and cellular response regulation.
