The CaMKII (Ca2+/calmodulin-dependent protein kinase II) family consists of four isoforms: CaMKII alpha CaMKII beta CaMKII delta and CaMKII gamma also known respectively as CAMK2A CAMK2B CAMK2D and CAMK2G. These are serine/threonine-specific protein kinases with a molecular weight of around 50 to 60 kDa. They play a role in the phosphorylation of various downstream targets. These kinases are found in many tissues but CaMKII alpha is mostly found in brain tissue CaMKII beta in muscle tissue whereas CaMKII delta and gamma show more widespread expression across other tissues. The functional unit of CaMKII is a holoenzyme which forms a dodecamer consisting of 12 subunits.
Biological function summary

CaMKII isoforms participate in critical calcium signaling processes. They act to decode the frequency of calcium oscillations in cells and are involved in synaptic plasticity and memory formation. CaMKII alpha for example is highly active in the postsynaptic density where it can form complexes with both NMDA and AMPA receptors suggesting a role in the regulation of synaptic strength. While not part of a permanent complex CaMKII enzymes can transiently bind to other proteins during signaling events.

Pathways

CaMKII isoforms play significant roles in the calcium/calmodulin signal transduction pathway and the MAPK (mitogen-activated protein kinase) signaling cascade. In the calcium/calmodulin pathway CaMKII acts to mediate calcium-dependent phosphorylation events that affect neuronal and cardiac functions. It interacts with other pathway proteins such as calmodulin and CREB (cAMP response element-binding protein). In the MAPK pathway CaMKII modulates signals that control cell growth differentiation and response to cellular stress interacting with proteins like MEK and ERK.

CaMKII isoforms have connections to neurodegenerative diseases such as Alzheimer's disease and to cardiac conditions like heart failure. In Alzheimer's disease altered CaMKII activity can affect synaptic function linking to proteins like amyloid-beta peptides. In cardiac disorders abnormal CaMKII activation contributes to disrupted calcium homeostasis and arrhythmia where it interacts with proteins such as RyR2 (ryanodine receptor 2). Understanding these interactions highlights CaMKII's significance in both brain and heart health.

Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca(2+)/calmodulin-binding and autophosphorylation, and is involved in dendritic spine and synapse formation, neuronal plasticity and regulation of sarcoplasmic reticulum Ca(2+) transport in skeletal muscle (PubMed : 16690701). In neurons, plays an essential structural role in the reorganization of the actin cytoskeleton during plasticity by binding and bundling actin filaments in a kinase-independent manner. This structural function is required for correct targeting of CaMK2A, which acts downstream of NMDAR to promote dendritic spine and synapse formation and maintain synaptic plasticity which enables long-term potentiation (LTP) and hippocampus-dependent learning. In developing hippocampal neurons, promotes arborization of the dendritic tree and in mature neurons, promotes dendritic remodeling. Also regulates the migration of developing neurons (PubMed : 29100089). Participates in the modulation of skeletal muscle function in response to exercise (PubMed : 16690701). In slow-twitch muscles, is involved in regulation of sarcoplasmic reticulum (SR) Ca(2+) transport and in fast-twitch muscle participates in the control of Ca(2+) release from the SR through phosphorylation of triadin, a ryanodine receptor-coupling factor, and phospholamban (PLN/PLB), an endogenous inhibitor of SERCA2A/ATP2A2. In response to interferon-gamma (IFN-gamma) stimulation, catalyzes phosphorylation of STAT1, stimulating the JAK-STAT signaling pathway (By similarity). Phosphorylates reticulophagy regulator RETREG1 at 'Ser-151' under endoplasmic reticulum stress conditions which enhances RETREG1 oligomerization and its membrane scission and reticulophagy activity (PubMed : 31930741).
