Flow Cytometry (Intracellular) - Anti-CaMKII alpha antibody [EPR1828] - BSA and Azide free (AB239906)
Overlay histogram showing SH-SY5Y cells stained with ab92332 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab92332, 1/1000 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in SH-SY5Y cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions. This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab92332).

Biological function summary
CaMKII alpha plays an important role in synaptic plasticity a process vital for learning and memory. The protein acts as an important component of the postsynaptic density complex where it interacts with other proteins to regulate synaptic strength. When calcium levels rise CaMKII alpha is activated through phosphorylation altering its conformation and allowing interactions with downstream substrates. This regulatory mechanism highlights its role in modulating neurotransmission synaptic potentiation and memory consolidation.
Pathways
CaMKII alpha functions within the calcium signaling pathway contributing to long-term potentiation (LTP) and long-term depression (LTD). It interacts with proteins such as NMDA receptors to facilitate changes in synaptic strength alongside other kinases. This pathway's activation leads to memory formation and synaptic plasticity in the central nervous system. Additionally CaMKII alpha connects with the MAPK signaling pathway integrating signals from various neurotransmitter systems which help coordinate neuronal response to stimuli.
