Supplementary information

CAPZA2 also known as capping protein (actin filament) muscle Z-line alpha 2 is a protein that mechanically functions to regulate actin filament dynamics by binding to the barbed ends. This protein has an approximate molecular mass of 33 kDa. CAPZA2 is predominantly expressed in muscle tissues suggesting its significant role in muscle function. The protein forms part of the actin capping protein complex specifically interacting with the CAPZB subunit.
Biological function summary

CAPZA2 controls the growth of actin filaments an essential process for cell motility and stability. It is part of the capping protein complex which is necessary for the capping and stabilization of the fast-growing ends of actin filaments. This stabilization prevents further polymerization maintaining optimal filament length for cellular dynamics. CAPZA2 also plays a role in cytoskeletal organization important for muscle contraction and cell shape maintenance.

Pathways

The actin-regulating activity of CAPZA2 integrates into pathways involving actin cytoskeleton organization and dynamics. It is significantly involved in the actin-mediated processes interacting with proteins such as Arp2/3 complex which initiates actin branching. CAPZA2's role in cytoskeleton assembly also positions it within the signaling pathways related to cell growth and muscle function impacting interactions with proteins like tropomyosin which stabilizes actin filaments.

CAPZA2 shows a connection to muscular disorders including myopathy. Dysfunction in actin filament regulation can lead to issues in muscle fiber integrity and function contributing to muscle weakness or degenerative conditions. CAPZA2 is also linked to cardiac disorders due to its important role in muscle contractility interacting with proteins like myosin which is vital for heart muscle function. Understanding CAPZA2's roles can aid in developing therapeutic targets for muscle-related diseases.

Target data

F-actin-capping proteins bind in a Ca(2+)-independent manner to the fast growing ends of actin filaments (barbed end) thereby blocking the exchange of subunits at these ends. Unlike other capping proteins (such as gelsolin and severin), these proteins do not sever actin filaments.
